Top Line: Is rheumatoid arthritis associated with a higher risk of radiation related toxicity?
The Study: RA is an autoimmune disease characterized by complex polygenic alterations. Many of the genes associated with RA are also involved with DNA damage response. This study analyzed germline genetic data from 3797 patients who received curative intent radiation for lung cancer, breast cancer, and prostate cancer. They calculated a polygenic risk score (PRS) using 101 genetic variants associated with RA. Overall, PRS was not significantly associated with the risk of developing either acute or late toxicity after RT. Among individual disease cohorts, a higher PRS (top 10th percentile) was associated with esophagitis in lung cancer patients, and it was associated with acute rectal bleeding and late hematuria in prostate cancer patients. However, these individual associations did not hold up in the validation cohort.
